What is Adafil?
Adafil® (20 mg tablet) is the preparation of Tadalafil. Tadalafil is a potent, selective, reversible inhibitor of cyclic guanosine monophosphate (cGMP)-specific phosphodiesterase type 5 (PDE5).Tadalafil is the only PDE5 inhibitor that works up to 36-hours due to its longer half life.
How Adafil works in our body?
Tadalafil enhances the effect of nitric oxide (NO) by inhibiting phosphodiesterase type 5 (PDE5), which is responsible for degradation of cyclic guanosine monophosphate (cGMP) in the corpus cavernosum. Tadalafil has no direct relaxant effect on isolated human corpus cavernosum and has no effect in the absence of sexual stimulation. When sexual stimulation causes local release of NO, inhibition of PDE5 by Tadalafil causes increased levels of cGMP in the corpus cavernosum, resulting in smooth muscle relaxation and inflow of blood to the corpus cavernosum.
What is the indication of Adafil?
Adafil is indicated in erectile dysfunction (ED).
It is also indicated in pulmonary arterial hypertension (PAH).
What are the dosage & administration of Adafil?
ED: The recommended starting dose of Tadalafil in most patients is 10 mg, taken prior to anticipated sexual activity (30 minutes before). The dose may be increased to 20 mg or decreased to 5 mg, based on individual efficacy and tolerability. The maximum recommended dosing frequency is once per day. Tadalafil may be taken without regard to food.
PAH: The dose for most patients is 40 mg, consisting of two 20-mg tablets taken once-daily. Both 20-mg tablets of Adafil should take at the same time of day every day. It can be taken with or without food.
What are the contraindications of Adafil?
Nitrates and Tadalafil must not be used concomitantly. Co-administration of Tadalafil with nitric oxide donors, organic nitrates or organic nitrites in any form either regularly or intermittently is contraindicated. Drugs which must not be used concomitantly include, but are not limited to, glyceryl trinitrate (injection, tablets, sprays or patches), isosorbide salts, sodium nitroprusside, amyl nitrite, nicorandil, or organic nitrates in any form. Tadalafil should not be used in patients with a known hypersensitivity to Tadalafil or to any of the excipients. Tadalafil is contraindicated in patients who have loss of vision in one eye because of non-arteritic anterior ischaemic optic neuropathy (NAION), regardless of whether this episode was in connection or not with previous PDE5 inhibitor exposure.
What are the adverse effects of Adafil?
Most common adverse reactions include-

• Dyspepsia
• Back pain
• Myalgia
• Nasal congestion
• Flushing and
• Pain in limb.

High risk group
Tadalafil is not indicated for use in newborns, children or women.
What would be the storage condition of Adafil?
Store at a cool & dry place, away from light. Keep out of reach of children.
